Research Abstract

BRCA1のユビキチンリガーゼ(E3)活性による乳癌抑制機序を解明するために本研究では基質であるヌクレオフォスミン(NPM1)とRPB8、またBRCA1の活性を制御する可能性を持つBAP1について解析した。それぞれ以下の結果を得た。NPM1(投稿中) : (1)Thr199リン酸化型NPM1(pT199NPM1)が電離放射線によるDNA二本鎖切断部位(DSB)に集積する。(2)この集積はRNF8,Ubc13,Lys63結合型ユビキチン鎖のDSBへの集積に依存している。(3)DSBにおけるpT199NPM1のturn overにBRCA1/RAP80が必要。(4)T199NPM1リン酸化を抑制するとDSBにおけるRAD51のturn overが阻害され、DNA損傷が助長され、細胞の電離放射線に対する感受性が亢進する。RPB8 : (1)BRCA1がRPB8をユビキチン化する際、まず異的なE2とともにモノユビキチン化する。(2)このE2をsiRNAにて抑制するとUV後のRPB8のポリユビキチン化が抑制される。(3)内因性RPB8の発現を抑制し、ユビキチン化されないLys/Arg変異体をadd-backするシステムを確立。BAP1(Cancer Res. 69 : 111-9, 2009) : (1)BAP1はBARD1のRINGフィンガードメインにBRCA1と競合的に結合する。(2)これによりBRCA1/BARD1のE3活性を阻害する。(3)E3活性阻害に加え、BRCA1/BARD1によって生じたポリユビキチン鎖を脱ユビキチン化する活性も併せ持つ。(4)BAP1抑制により細胞のS樹が延長し、電離放射線に対する感受性が亢進する。
