Research Abstract

アポトーシスや細胞膜抗酸化作用との関連を追究するために、プリオン及びドッペルの発現ベクターを作製して、リンパ系細胞に強制発現させ、紫外線照射時や血清除去時のアポトーシスに対する細胞の挙動変化を解析した。プリオン関連蛋白質と微小RNAとの結合の有無を検討したが、特定のRNAとは結合していない。ドッペル蛋白質ではプリオン蛋白質と異なる機能が報告されており、Bcl2関連アポトーシスカスケードの中で機能していることが示唆された。アダプター蛋白質NESHとの相互作用を免疫沈降やウエスタン法で確認した。SOD活性については、WAT1を用いて測定し、ドッペル蛋白質がSOD活性を阻害していることが判明した。
