Research Abstract |
銅排出酵素系において特異的にCu(I)を酸化する酵素CueOから50残基のアミノ酸を除去し,有機基質に対する酸化活性を創製するとともに,触媒部位への配位アミノ酸への変異導入や水素結合の切断や形成によって、触媒機能を高めることに成功した。また,CueOの組換え体および変異体を,生物燃料電池のカソード触媒やヘアカラー色素作成酵素としての利用することを可能とした。さらに,変異体の反応によって,酸素の4電子還元機構を明らかにし,反応に関与する非配位性の酸性アミノ酸を特定した。
