Research Abstract

mRNAディスプレイ(IVV)法を用いて、合成一本鎖抗体(scFv)ライブラリーから濃縮効率100万倍以上の高効率で、様々な抗原に結合するscFvや、on-rate選択およびoff-rate選択により親和性の向上したscFvを迅速かつ簡便に取得する手法を確立した。また、同様の手法を単一のドメイン抗体(ナノ抗体)の試験管内進化にも適用できた。次に、DNAディスプレイ(STABLE)法を用いて、二本鎖のFab抗体の試験管内選択に世界で初めて成功した。さらに、STABLE法のスクリーニング効率を上げるために、ビーズエマルジョンPCRとSTABLE法とを組み合わせた新しい手法を開発した。
