Research Abstract

スフィンゴ脂質は、形質膜ミクロドメインに主として存在し、細胞内のシグナリングや細胞間の相互作用に重要な役割を演じている。本研究では、申請者らが発見した非リソソーム性スフィンゴ脂質代謝酵素の構造と機能について研究を行い、以下のような成果を上げた。先ず、哺乳動物の細胞質に存在するグルコシルセラミド(GlcCer)分解酵素(エキソ型グリコセラミダーゼ)をクローニングした。本酵素は、老化関連タンパク質クロトーのホモログ(KLrP)であった。本酵素の活性をRNAi で抑制すると、細胞内のGlcCer 量およびGlcCer骨格を持つ糖脂質量が減少することを見出し、KLrPが細胞の糖脂質代謝に関わっていることを示した。本酵素を大腸菌で大量に発現させ、グルコース及びガラクトースとの共結晶を作製し、X線結晶構造解析を行った。その結果、KLrPはαヘリックスとβストランドを8回繰り返す、典型的なTIMバレル構造を示すこと、β-ストランド4および7のC 末に存在するE165とE373がそれぞれ酸/塩基触媒残基、求核残基であることを証明した。また、本研究においてリソソーム性の中性セラミダーゼの結晶構造解析に初めて成功した。その構造は今までに報告のない新しいものであり、その触媒機構は亜鉛を触媒中心とすることを明らかにした。構造を解くことで、スフィンゴ脂質シグナリングや食餌由来のセラミドの代謝・消化への関与等、本酵素の多彩な機能が合理的に説明できた。一方、放線菌から単離したエンド型グリコセラミダーゼ(EGALC)の基質特異性を詳細に調べ、本酵素は6ガラ系列糖脂質に特異的に作用することを明らかにした。また、EGALC は加水分解のみならず糖鎖の転移反応も効率よく触媒することも明らかになった。この特異性を利用した、6-ガラ系列糖脂質の蛍光標識法及び微量定量法を開発し、その方法を用いて、病原性の原虫、糸状菌、及び食用海藻等に6-ガラ系列糖脂質が存在することを見出した。海藻から単離した6-ガラ系列糖脂質の構造解析を行い、その構造を1-phytyl-3-O-[α-D-galactopyranosyl-(1-6)-β-D-galactopyranosyl]- glycerol と同定した。このような構造の糖脂質は初めての報告である。
