Research Abstract

薬物誘発性の遅延性臓器障害の機序解析のため、胆汁鬱滞型肝障害を誘発するlithocholicacid(LCA)とヒト特異的に肝障害の認められる前立腺癌治療薬のflutamideをモデル化合物として解析した。LCA誘発肝障害は肝内脂肪酸、トリグリセリド、リン脂質のレベルを増加させることにより軽減されることを明らかにした。Flutamide誘発肝障害に関わる代謝活性化体(FLU-1 N-OH 体)を同定し、その生成と分解の機序を明らかにした。これによりflutamideにより肝障害を起こし易いヒトを予測出来る可能性が示された。
