Research Abstract

発生・再生現象におけるSUMOタンパク質修飾機構と細胞核内構造のリモデリングを解析し、タンパク質修飾機構の意義を示すとともに、遺伝情報制御の基本メカニズムを解明することを目的とした。(1) メチル化DNA 結合タンパク質MBD1とポリコームタンパク質群の協働性の解析 : DNAメチル化とヒストンH3の9番目のリジン(H3K9)のメチル化を介するMBD1複合体経路、ヒストンH3の27番目のリジン(H3K27)のメチル化を介するポリコーム複合体経路が発生分化に働く遺伝子制御に関わっている。MBD1の結合因子を探索し、ポリコームタンパク質群(Ring1b、hPc2)と相互作用することを見出した。両経路が直接的に相互作用することで、HOXA 遺伝子群の転写抑制とヘテロクロマチン形成に協働することが示唆された。(2) 細胞周期におけるポリコームタンパク質群の解析 : ポリコーム群にはPRC1とPRC2の2つの複合体があり、PRC2(メチル化酵素EZH2を含む)はH3K27をメチル化し、PRC1(SUMOリガーゼhPc2を含む)はメチル化H3K27を認識し転写抑制型のクロマチンを形成する。PRC1が結合したクロマチンが細胞周期のG1期に再構築され、次のS期への進行に必要であることを見出した。細胞がG1期に刺激を受けた場合、PcGの標的クロマチンが再編される可能性を示唆した。(3) SUMO結合モチーフをもつMCAF1の解析 : ユビキチン様SUMOで修飾されたMBD1がMCAF1分子内のSUMO結合モチーフと相互作用することを報告したが、新たにMCAF1が転写因子Sp1に直接結合し、Sp1依存性のテロメラーゼ遺伝子発現を促進することを見出した。MCAF1が相互作用する標的タンパク質のSUMO修飾が異なる複合体を形成する可能性を示唆した。(4) SUMO修飾が関わる核内構造の解析 : 核膜孔に存在するSUMOリガーゼRanBP2が、SUMO修飾と関連する核内構造、とくにPMLボディーや上記のヘテロクロマチン等の形成に関わることを見出した。このように、タンパク質のSUMO修飾機構が、遺伝子発現と核内構造体の形成に重要な役割を果たすことを明らかにした。
