Research Abstract

ストレス応答MAPK経路(MAPKKK-MAPKK-MAPK)は、環境ストレスによって活性化され、細胞のストレス応答の制御に重要な役割を果たしている。また、この経路の制御異常が癌や慢性炎症性疾患に深く関与する。本研究では、ストレス応答経路の主要なヒトMAPKKKであるMTK1の活性制御機構の解明を行った。その結果、1)ストレス刺激後にGADD45分子の発現が誘導されてMTK1と結合すると、まずMTK1制御ドメインと酵素ドメイン間の抑制的な分子内相互作用が解除されてMTK1が多量体化し、さらに隣接したMTK1分子同士が相互にリン酸化し合って、強い活性化が起こることを見出した。また、2)MTK1の新たな活性化促進分子としてRACK1を同定した。さらに、低酸素によってストレス顆粒が形成されると、RACK1が顆粒内に取り込まれてMTK1-p38/JNK経路が失活し、DNA損傷による細胞死が阻害されることを見出した。即ち、ストレス顆粒形成によるMTK1の活性化阻害が、腫瘍内部低酸素環境において癌細胞が抗癌剤抵抗性を獲得する一因となっていることを示した。
