Research Abstract

咽頭や口腔粘膜での病原性細菌の病原性の発揮には,宿主組織への付着により生体組織に定着することから開始する.そのため,生体を広く覆う上皮や粘膜といった組織は,これらの菌の侵入を感知する最前線の組織であり,かつ最大の防御組織となっている.近年,多くの細菌種がこのような上皮組織の細胞内に侵入することが知られているが,その動態はほとんど明らかとされていない.特に,貪食系の細胞ではない上皮細胞内に取り込まれた菌が,エンドソーム・リソソームの融合システムでのみ効率的に分解されるのか否かについてはほとんど明らかとされていなかった.申請者は,宿主細胞が細胞内の菌体成分認識分子であるNalp のある分子が、細胞質内に侵入した菌の認識に重要で、かつ炎症反応の惹起の切り替えにこのシステムが機能していることを見いだした.通常の菌体成分の認識機構であるNod2 は炎症反応の惹起に重要であるが、Nod1, Nod2 そのものはオートファジーの誘導には必須ではなく、炎症反応を惹起しないNalp4, Nalp10 がオートファジーの誘導に必須であることを明らかとした.
