Research Abstract

札幌医大および順天堂大学で開発した独自の癌標的化抗体作成法を通じて樹立に成功した新規腺癌抗原PAP2aおよび数種の癌関連抗原に対する多くの高性能抗体を用いて癌の高感度かつ低侵襲性の診断法の開発を目指した。1)大腸癌検診の精度を高めるために癌標的抗原を便中から高感度で検出する検査方法の開発を進めた。これまでに当該研究で樹立した高性能がん標的抗体を用いて確立できたELISA系は5種類である。その中でも特に大腸癌と関連のある標的分子の測定を行った結果、癌関連プロテアーゼ誘導分子CD147および胎児性癌抗原CEAは健常人、大腸癌患者由来の便中では全く差が認められなかった。それに対して癌抗原XXXXX(特許出願準備中のため抗原名省略)は大腸癌患者由来便中に検出され、その値は健常人由来便中の抗原量よりも有為に高いことが示された。この結果をもとに現在、国内診断薬開発企業と金コロイド粒子法を用いた簡便迅速な大腸癌検査キットの開発を進めている。2)膀胱癌の早期診断のために癌標的抗原を高感度で検出する検査方法の開発を進めた。3種類の癌標的分子の測定を行った結果、尿中の上皮癌抗原EpCAMおよびCEA値は健常人、膀胱癌患者間で全く差がなかった。それに対して癌抗原YYYY(特許出願準備中)値は健常人由来尿中の抗原量よりも明らかに高いことが示された。この結果をもとに現在、国内診断薬開発企業と共同でイムノクロマト法を用いた簡便迅速な検査キットの開発を進めている。
