Research Abstract |
初年度において、各種リコンビナントコレクチン作成とその抗体作成を行った。遺伝子改変動物の作成については、ノックアウトマウスはCL-P1、CL-K1について両者とも、キメラマウスが樹立でき、さらにヘテロマウス作成が終了し、精子保存にてノックアウト表現型解析に進んでいる。平成22年度末の時点で、CL-P1は、仔マウスとして誕生せず、胎生致死に関与する遺伝子であると考えられた。一方、CL-K1ノックアウトマウスは無事誕生し、一見正常に見えた。しかしながら、ノックアウトマウス個体が野生型の通常マウスに比較して小型であることや、誕生個体が少ないことより、個体発生や生育に関与する遺伝子である可能性がでている。腎組織おける検討では、局在は異なるが両コレクチンの存在することが、本研究により再確認されたが、上記マウスを用いた病態における意義については十分な解析結果が得られていない、今後の検討が待たれる。
