Research Abstract

本研究課題においては、転写因子Fox01が膵細胞の分化や膵細胞型の決定に果たす役割をvivoで明らかにする為に、膵特異的Fox01欠損マウス(KO)と膵特異的Fox01トランスジェニックマウス(TG)を作成し、解析を行った。耐糖能はKOで優位に改善、逆にTGでは悪化し、一部のマウスは顕性の糖尿病を発症した。重要なことに、KOではインスリン陽性の膵管細胞と小さなサイズのラ氏島の増加が認められ、・細胞の新生が促進されていると考えられた。一方、TGでは明らかな・細胞の減少とともに、腺房細胞の著減、膵管の増加とラ氏島内血管の増加が認められた。これらの結果から、Fox01が膵細胞の分化、新生のみならず、その機能にも重要であることが明らかとなった。
