| Research Abstract |
新規遺伝子変異検索技術SMAP法を用い、臨床検体を用いたEGFR及びK-ras変異検出の検証、パラフィン切片からの検出、摘出組織をそのまま鋳型とした検出系を検討した。またEGFR変異検出システムの改良を行った。結果、検出限界は1%未満であり、パラフィン切片からの診断もほぼ全例で可能であった。摘出組織をそのまま鋳型とした検出では、摘出から約1時間という超迅速診断の開発に成功した。EGFR exon19変異検出においてはPNAを応用することでほぼ全ての変異型を検出可能にした。以上のことより、SMAP法は臨床現場での遺伝子診断に有用であることが確認された。
