Research Abstract

ヒストンの脱メチル化による遺伝子発現制御機構を解明する目的で、それを触媒する酵素の機能解析を行った。ヒストン脱メチル化酵素、JHDM2はA, B, Cの3種があり、それぞれについてノックアウトマウスの表現型を調べた。そのうちで、JHDM2A遺伝子改変マウス雄は不妊の表現型を示した。雄の不妊の原因をさらに詳細に調べた結果、減数分裂期のヒストンH3のリジン9のメチル化が亢進していた。この事実によって、雄特異的な減数分裂期の積極的な脱メチル化反応がJHDM2Aによって触媒されることが明らかとなった。
