Research Abstract

ミクロとセミミクロカラム用の液体クロマトグラフ(HPLC)システムで、確立した精製法で、つぎの標品を使用してLC-MSシステムにより網羅的な分析を行った。対象は、内部標準として必須の安定同位体ラベルの標品が入手可能なホルモンである ; アブシジン酸(ABA)、インドール酢酸(IAA)、ジベレリン類(Gas)はGA1・GA4・GA19・GA20・GA53、サイトカイニン類(CKs)はゼアチン(Z)・リボシルゼアチン(ZR)・イソペンテニルアデニン(iP)・イソペンテニルアデノシン(iPR)、ジャスモン酸類(Jas)はジャスモン酸(JA)・メチルジャスモン酸(MJA)。特にJasは、揮発性が高いので工夫を要した。キュウリ果実を材料として、アルカリの加水分解処理により配糖体からフリーのホルモンを遊離させて、抽出・精製後にLC-MSで定量する方法で、ABA・IAA・Gas・CKsの配糖体を定量した。キュウリ果実を細かく組織に分けて、アポプラスト(AP)液とシンプラスト(SP)液を遠心法で採取した。これらの画分に安定同位体標識試薬を内部標準として加えて、確立した分析法で、植物ホルモンおよびそれらの配糖体の局在性および消長を解析した。内生ホルモンの合成部位や貯蔵器官の特定のため、免疫組織化学染色法により、植物ホルモンのIAA, ABAおよびサイトカイニンのイメージング(可視化)技術を確立して、キュウリのつぼみから幼果までの解析を行った
