Research Abstract |
葉緑体NDH複合体の未知サブユニットを同定する目的で、Arabidopsis共発現解析データベース等を利用して新規NDH遺伝子の選抜をした。その結果、5個の遺伝子の同定と解析をライバルに先駆けて、または同時に発表することができた。 C_4種、C_3種および intermediate のフラベリアを材料とし、4個のNDH サブユニットに対する抗体を用いてイムノブロット解析を行った。また、C_4フラベリア(F. bidenti)から、シロイヌナズナで同定された核ゲノムコードのNDHサブユニット遺伝子ndhNと本研究にて単離したndf4, ndf6のホモログをクローニングした。単離した遺伝子配列を利用してその遺伝子の発現抑制を引き起こすためのRNAiベクターを作成し、形質転換を行った。
