Research Abstract

本研究ではIRSと結合するDGKζ活性がGlut4 の細胞膜移行を阻害するメカニズムを解明し、インスリン抵抗性の発生とIRS-1 結合性DGKζとの関連を明らかにすることを目的としている。3T3-L1 adipocytes において、siRNA によってDKGζをノックダウンすると、インスリン非依存的な糖取り込み量に変化は認められないが、インスリン依存的な糖取り込み量が有意に減少すること、さらにインスリン依存的なインスリンシグナル分子の活性化には全く影響しないことを明らかにした。続いて、阻害剤を用いて、DGK の活性を阻害したところ、インスリン依存的な糖取り込みが促進されることが明らかとなった。しかし、インスリン依存的なインスリンシグナル分子の活性化には全く影響を与えなかった。一方で、糖取り込み担体であるGLUT4 の細胞膜移行はインスリン非依存的に促進されており、DGK 活性はGLUT4 の細胞膜移行を阻害していることが明らかとなった。これらの結果から、DGKζは、複雑な機構によってこれまでに知られているインスリンシグナルとは異なる新規シグナルを介して、インスリン依存的な糖取り込みを制御していることが明らかとなった。
