Research Abstract

[1] 腫瘍壊死因子関連タンパク質の一つで細胞死を誘導する"TRAIL"が結合する, 膜受容体タンパク質"DR5"は, TRAIL耐性の腫瘍細胞では核内に格納され, その結果, TRAILにより誘導される細胞死の脅威から逃れている可能性があることが示唆された. TRAIL耐性の腫瘍細胞の核内DR5を, 細胞膜へ発現させることで, 細胞死を誘導できることが明らかになった.
[2] 胆汁うっ滞性疾患において, 胆管上皮細胞上のTRAIL/DR5を介して, 重篤な胆管炎と肝障害が引き起こされる危険性があり, TRAIL/DR5を標的としたヒトへのがん抗体療法を行う上で, 副作用に注意する必要性が示された.
