Research Abstract

血小板のコラーゲンレセプターGPVIの特異的な阻害剤の探索及びそれを用いた反応機構の解析を行った。
1. GPVIダイマーと特異的に結合するFabをスクリーニングし、コラーゲンとの結合にはGPVIのダイマー構造が必要なことと、血小板上でGPVIはダイマーとして存在することを示した。
2. GPVIと複合体を形成するFcRγ鎖の透過性ペプチドを作製した。このペプチドはコラーゲンを介した血小板の活性化を特異的に阻害し、特にSykのリン酸化を阻害した。
3. 変異GPVIの解析から、R38とE40およびR65が結合活性に強く関与していることを示した。
