Research Abstract

ヒト血液凝固内因系・高分子キニノーゲンの分解産物であるドメイン5(D5_H)の持つ細胞の接着阻害, 走化阻害, 浸潤阻害とその阻害活性機構, 内皮細胞管腔形成阻害, アポトーシス誘導活性を検討した。(1)細胞の種類と誘因タンパク質(細胞外基質タンパク質)の種類にかかわらず, D5_HおよびD5_H由来ペプチドによる細胞の接着阻害, 走化阻害, 浸潤阻害が見られた。(2)阻害活性を発揮するD5_H の領域に結合する細胞膜画分を同定した。 (3) 内皮細胞管腔形成阻害活性を持つと思われる領域を同定した。(4)アポトーシス誘導活性は細胞接着阻害実験後の細胞において認められなかった。(5)抗体を用いた細胞染色法によるERK1/(2)とAktのリン酸化の度合の変化は, 細胞接着阻害実験後の細胞では見られなかった。
