Research Abstract

野生型に比べSTAM1/2二重欠損マウス由来の樹状細胞においては、抗原提示能の増強ならびにMHC classIIの細胞表面での発現の亢進が見られた。mRNA測定ならび蛋白分解実験からMHC classIIの発現増強は蛋白合成の亢進によるものではなく、蛋白質分解の抑制によるものであることが明らかとなった。また、電子顕微鏡による観察から、STAM1/2二重欠損樹状細胞ではMHCII貯蔵小胞であるMVBの形成異常みられ、MHC classIIの分解異常を指示するデータが得られた。以上のことから、STAM1/2は、MHC classII の分解を制御することにより、MHC classIIを介する抗原提示に重要な機能を果たす分子であることが明らかとなった。
