| Research Abstract |
生体内生理活性物質の正確な測定は臨床化学の基盤であるが, 低分子化合物 (ハプテン) の高感度測定は高分子化合物に比べて難しい. 新しい分子認識単位として, "ナノ抗体", すなわち,単一ドメイン抗体 (single-domain antibody; sdAb) を導入することで, ハプテンの高感度モニタリングが可能と期待される. SdAb とは,ラクダの "H 鎖抗体" の可変部ドメイン (VHH) あるいはそれを模倣したタンパク質で, 抗原結合能を保持する最小の抗体フラグメントである. 以上の観点から, ステロイドをモデルハプテンとしてマウス抗体の可変部ドメイン (VH またはVL)を基本構造とするsdAb を遺伝子工学的に作成し, そのハプテン結合能を検討した.
