Research Abstract |
一酸化炭素中毒時には、活性酸素種の中で最も毒性の強いヒドロキシルラジカルの生成が脳内で増加するため、これが一酸化炭素中毒による脳障害に関与することが推察されている。本研究において、一酸化炭素中毒時には、脳内の細胞外アスコルビン酸が増加し、これが遊離鉄と反応してヒドロキシルラジカル生成の増加の機序の一つであることが示唆され、通常は脳内で抗酸化剤としての役割を担うと考えられているアスコルビン酸が、一酸化炭素中毒時には、プロオキシダントとしての作用を発揮することが示された。このようなアスコルビン酸の作用は、デヒドロアスコルビン酸を腹腔内投与して脳内の細胞外アスコルビン酸を増加させたモデルでも再現されたことにより裏付けられたことから、in vitro では古くから知られていたアスコルビン酸のプロオキシダントとしての作用が、ある条件下ではin vivo においても発現しうることが示された。
