Research Abstract

C型肝炎ウイルス(HCV)コホート研究にて、コレステロール生合成を制御するTM7SF2遺伝子の多型として433番アミノ酸のThrからIleへの置換が、HCV 感染成立に影響する遺伝要因の一つであることを見出した。TM7SF2のHCV増殖への関与は、TM7SF2 siRNAによりTM7SF2ノックダウン細胞を作成し、HCVレプリコンを導入後にHCV RNA発現量を測定したところ、HCV RNA の細胞内発現量が、ノックダウン細胞では、非ノックダウンル細胞のそれと比較して低下することで証明した。
