Research Abstract

本研究の目的はC 型肝炎ウイルス(HCV)増殖制御因子として申請者らが新たに明らかにした非構造蛋白NS3ヘリカーゼ・ドメインに着目し、同領域によるHCV増殖制御機構の解明を通して、新たな抗HCV治療戦略の基盤を形成することにある。申請者らは本研究において、HCVゲノムの相同組み換え実験により、ウイルスゲノム増殖にはHCV の非構造蛋白NS3ヘリカーゼドメインが決定的な役割を果たすことをまず明らかとしたが、詳細なマッピングを行うことにより、さらに最終的に6アミノ酸部位に絞られることを示した。一方、ウイルス増殖を制御する宿主の主要機構である自然免疫との関連に注目し、RIG-Iに誘導されるインターフェロン発現シグナルをNS3 蛋白は構造依存的に抑制することを明らかとした。
