Research Abstract

健常成人のボランティアより採取した末梢血より比重遠心分離法により単核球を分離し、さらにCD4 T細胞分離カラムを用いてCD4 T細胞を分離し、Anti-human CD3 mAbをcoatingしたplateでIL-23存在下に培養し、IL-17の産生を特徴とするヒトTh17細胞を誘導した。Th17細胞を非小細胞肺癌細胞や肺線維芽細胞とトランスウエルを用いて48時間共培養し、培養中の多種類の血管新生因子の濃度をELISA kitを使用して測定した。Th17細胞を非小細胞肺癌細胞や肺線維芽細胞とトランスウエルを用いて共培養すると、肺癌細胞や線維芽細胞からの血管新生因子IL-8、ENA-78、GRO-alphaなどの産生が著明に増加した。この現象は、樹状細胞などより分泌されるIL-12で誘導されるTh1細胞と、非小細胞肺癌細胞や肺線維芽細胞を共培養すると、培養液中の血管新生抑制因子IP-10やMIGの濃度が、Th1細胞や肺癌細胞を単独で培養した際の培養液中の濃度に比較して著明に上昇することと極めて対照的であった。
