Research Abstract

本研究において, われわれは以下の点を明らかにした.
(1)ラット脳塞栓組織型プラスミノゲン・アクチベーター(tPA)投与モデルを確立し, tPA療法後の脳出血抑制を目的とする新規治療薬のスクリーニングが可能となった.
(2)上記モデルにて, 治療可能時間(3時間)を過ぎてtPA療法を行った場合, 血管内皮細胞において高度のVEGF発現が生じていることを初めて明らかにした.
(3)tPAと同時に抗VEGF抗体を投与することで, 血管透過性を亢進し, 血液脳関門を破綻させるVEGF発現の抑制が生じたことから, VEGF発現の抑制は, tPA療法の欠点を改善する手段として有望である可能性がある.
