Research Abstract

α-ジストログリカンのプロッセッシングを解析するためにN末側およびC末側のコア蛋白質に対する特異的抗体を作製した。これらの抗体を用いて培養細胞における検討を行なったところ、α-ジストログリカンのN末端ドメインはプロプロテインコンバターゼによって切断された後ただちに細胞外へ分泌されることを明らかにした。さらにヒトの血清や髄液中にもこのα-ジストログリカンのN末端断片が存在することを示した。
悪性腫瘍由来の培養細胞には分子量75kD前後の低分子量のα-ジストログリカンが発現していた。これらのα-ジストログリカンはラミニン結合能を持たないことから異常な糖鎖修飾を受けたα-ジストログリカンと考えられた。培養細胞に対してLarge遺伝子のトランスフェクションを行なうと、α-ジストログリカンの分子量は200kD前後にまで著明に増大するとともに、ラミニン結合能は著しく亢進した。このように培養細胞にLargeを過剰発現させることによりα-ジストログリカンの機能修復が生じることを示した。
Largeを過剰発現するトランスジェニックマウスを作出した。同マウスは正常に誕生、発育し、交配も可能であり、外観上明らかな行動異常を示さなかった。同マウスの各臓器においてα-ジストログリカンの高分子量化とIIH6に対する反応性の著明な亢進を認めた。同マウスは明らかな障害を示さないことから、将来的にはLargeの全身的投与によるα-ジストログリカノパチーに対する治療への道が開かれたものと考えられる。
