Research Abstract

p85α欠損マウスの肝臓でのインスリン作用と糖代謝を比較検討した。グルコースクランプ法では、野生型と比較し、p85α欠損マウスの末梢組織でのインスリン感受性は亢進していたが、p85α欠損マウスの肝糖放出は抑制されていなかった。野生型と比較し、p85α欠損マウスの肝糖新生系酵素の遺伝子発現は上昇していた。また、肝臓でのAkt活性、PTEN活性は対照の野生型マウスと同程度であった。これらの結果より、p85α欠損マウスでは、末梢組織とは異なり、肝臓でのインスリン感受性は亢進しておらず、むしろ糖新生が亢進していると考えられた
