Research Abstract |
本研究は当研究チームで発見した新規2型糖尿病疾患感受性遺伝子TFAP2B、KLF7の2型糖尿病発症に関わる分子機構の解明を目的とした。ハムスター由来のインスリン分泌細胞(HIT-T15)を用いてインスリン分泌を検討したところ、TFAP2B,KLF7ともグルコースによるインスリン分泌を著明に抑制した。このTFAP2Bのインスリン分泌抑制効果は一部GLUT2, Kir6.2, HNF4A発現減少を介している事も明らかとなった。さらにTFAP2Bの膵β細胞特異的トランスジェニックマウスを作成し解析を開始しているが現時点では耐糖能異常等の表現形は認められていない。
