Research Abstract

VWFマルチマーを分解するADAMTS13の欠乏はマルチマーのダウンサイジング機構破綻=VWF機能の過剰をきたすと考えられる.ADAMTS13ノックアウトマウス をvon Willebrand因子(VWF)血中濃度が高いCAST/Eiマウスとの交配により血栓性血小板減少性紫斑病(TTP)様の血栓症を自然発症させ、我々が見いだしたがマウスVWFに交差反応する抗ヒトVWFモノクローナル抗体5D2をこれに直接投与、病態の修飾を観察し、「VWF機能亢進症」であるTMAの抗VWF抗体による病態制御の基盤的な知見を得た.
