Research Abstract

成人急性骨髄性白血病(AML)90例の白血病細胞のRhoH発現量をTaqMan PCR法で定量した.RhoH発現はFAB分類、年齢、初診時白血球数、寛解導入率、染色体異常の有無に無関係であった.RhoH低発現群と高発現群に分けて予後を検討したところ、低発現群では有意に予後不良であった.また多変量解析にて、RhoH低発現は無病生存率において予後不良因子であった.COS7細胞にRhoHを過剰発現させた状態ではsmall GTPase Racが不活性化することが明らかになった.またRac-PAKの下流に存在するBadのS112のリン酸化を測定したところ、RhoH発現によりBad S112のリン酸化が抑制される事が明らかになった.
