Research Abstract

自己免疫疾患、特に全身性エリテマトーデス(SLE)の病態における抗HMGB1抗体の役割を明らかにする目的で、血清中の抗HMGB1抗体のエピトープの解析を行い、その特徴と疾患活動性との関連を調べた。SLEにおいて抗HMGB1抗体が高頻度に検出され、抗HMGB1抗体とSLEDAIとの相関を認めた。エピトープ解析では抗HMGB1抗体は親水性で機能的な領域であるboxAおよびjoiner領域を主に認識した。また、joiner領域中の配列AKGKPDに様々な病原微生物の蛋白と分子相同性を認めた。一方、HMGB1分子はSLEや筋炎、成人Still病などの自己免疫疾患や顕微鏡的多発血管炎(MPA)、Wegener肉芽腫(WG)の患者血清で上昇を認め、HMGB1が自己免疫の病態に関与していることが示唆された。
