Research Abstract

これまでの研究によって、神経芽腫におけるMYCNの新規ターゲット検索を行った結果、遺伝子発現の抑制を介して発がん起序に深く関与するポリコーム複合体の構成分子BMI1を見出した。
MYCNによってBMI1の転写が調節され、In vivoにおけるMYCNのBMI1プロモーターへの結合をChIPアッセイで検証した。
BMI1の発現は神経芽腫細胞の増殖を促進した。また神経芽腫細胞の分化誘導においてBMI1が分化を抑制していることが示された。
さらに、BMI1のノックダウン系および過剰発現系の実験を行った結果、神経芽腫細胞の増殖・分化に関るBMI1の発現調節標的分子として、がん抑制遺伝子p14ARFとp16INK4a以外の重要な分子の存在が推定された。
このBMI1標的遺伝子のスクリーニングをBMI1ノックダウン細胞での発現チップ解析にて行った結果、当研究所でこれまで神経芽腫で予後を左右する因子であり、がん抑制遺伝子であると報告していた分子が標的である可能性を見出した。
