Research Abstract

Yin Yang-1(以下YY-1)は偏在する転写因子であり、様々な転写制御に関わっていると言われています。YY-1蛋白は多くの組織や細胞で同定されており、またその働きは組織や細胞で異なります。我々は以前YY-1蛋白が表皮基底層で強く発現され、上層に行くにつれて弱くなることを確認しました。この結果を元に、YY-1が表皮角化細胞の分化過程により重要な役割を担っているのではないかと考え、新たに3D培養法という手法と、角化細胞のセルラインであるHaCaT細胞を用い、研究を行いました。結果として、過発現したYY-1は3D培養において、表皮の著明な肥厚、分化の抑制、未分化状態の維持、及び分裂能の増加などを示しました。これらは、通常の皮膚組織において、YY-1が表皮基底層における角化細胞の未分化状態の維持と、上層での分化移行に重要な働きを示唆しています。
