Research Abstract

MRSにより測定されたグルタミン酸・グルタミン(Glx)とGABAの脳内濃度を統合失調症の中間表現型とみなし、これに影響を与える遺伝子の一塩基変異(SNP)を探索するために、ドパミンD3受容体(DRD3)、脳由来神経栄養因子(BDNF)、ニューレグリン1(NRG1)の遺伝子の計8つのSNPを調べた。同時に脳局所体積と脳白質拡散異方性も測定した。結果、前部帯状回、後部帯状回、小脳のN-アセチルアスパラギン酸、コリン含有物、Glx、GABA濃度は野生型と非野生型で有意差はなかった。脳局所体積についても同様であった。しかし脳白質の拡散異方性指標fractional anisotropy(FA)については、NRG1遺伝子のSNPの一つrs39249993の非野生型では右前頭連合野白質のFA値が有意に低かった。またこの変異を持つ統合失調症患者の右前頭連合野白質のFA値は、さらに低値であった。
