Research Abstract

鹿児島大学で開発されたクラウン系ミニブタ(近交系)の遺伝子改変(ジーン・ターゲティング)を行うために、ゲノムDNAより候補遺伝子を単離し、ターゲティング・コンストラクトを作製、ミニブタ胎児細胞に導入して検討した。遺伝子導入後、PCR産物のサザンブロットでは候補細胞が存在したが、ジェノミックサザンでさらに確認したところ目的とする細胞はないと判断された。この結果に基づいて、コンストラクトおよびターゲティングアリル検出方法の再検討をおこなった。
