Research Abstract

大腸癌手術検体のDNAチップ解析から我々が同定したKU-CR4は、高頻度かつ早期から大腸癌特異的に発現亢進がみられ、しかも細胞膜型と分泌型が存在することから、新規早期診断マーカー・治療標的として有望な分子である。本研究では、本遺伝子が大腸癌で発現亢進する機構、診断・治療への適用の可能性を検討した。抗体を複数作成しELISA検出系の構築を試みた結果、膜型蛋白質に対する抗体を用いたサンドイッチELISAによって大腸癌患者血清中にKU-CR4蛋白質を検出できた。この結果は血中のKU-CR4蛋白質の検出が大腸癌診断に応用できる可能性を示唆する。
