Research Abstract |
骨肉腫の治療成績は飛躍的に進歩し、専門施設における5年生存率は約70%まで向上してきた。しかし最近10年間での進歩はなく、残る30%は症例の既存の治療法に対して抵抗性である。さらに初診時にすでに遠隔転移を生じている症例(以後M1症例)の予後は著しく不良であり、5年生存率は10-40%にまで低下する。この見地から、これまでM1症例の生物学的特徴について、ゲノムレベルのいくつかの研究がなされているが未だ共通した特質は明らかにされていない。本研究では近年発生、癌化、分化へ深く関与していることが明らかにされつつあるDNAメチル化に着目し、これまで数個の遺伝子について断片的に行われてきた解析を、ゲノム全体のメチル化プロファイル(以下メチロームと称する)の解析を通して、骨肉腫M1症例の特質を解明することを行った。
