Research Abstract

肝細胞増殖因子トランスジェニックマウスと癌抑制遺伝子であるArf、Ink4a、p53それぞれのノックアウトマウスのダブルマウスを作成し、発生したメラノーマ、横紋筋肉腫を解析した。その結果、Arf-p53経路の不活性化と横紋筋肉腫発癌との関連性を見いだした。一方で、p53の不活性化とメラノーマ発癌とは関連を認めなかったのに対して、Arfの不活性化のメラノーマ発癌への関与が示された。このことはArfがp53を介しない経路でメラノーマ発癌の抑制をしている可能性を示唆した。Ink4a-RB経路の不活性化はメラノーマ発癌に関与する一方で、横紋筋肉腫発癌への関与は認めなかった。
