Research Abstract

破骨細胞は造血幹細胞に由来し、骨微小環境下に形成された単核の破骨前駆細胞どうしが融合することにより大型で多核の破骨細胞が形成される。更に骨芽細胞や骨基質から提供される刺激を受け、骨吸収機能に必須の形態学的超構造であるruffled border(刷子縁)が形成される(しばしば「活性化」という言葉で表現される)と、破骨細胞の活発な骨吸収が始まる。破骨細胞分化因子RANKLとその受容体であるRANKの発見により、分化シグナルに関する知見は急速に集積されつつあるが大規模な形態学的変化を伴う破骨細胞の「活性化」とその制御機構に関しては不明の点が多い。本研究では、破骨細胞の活性化に伴う特異的膜タンパク質の同定と破骨細胞の機能発現に伴う形態学的な動態解析を行い破骨細胞活性化の制御機構の一端を解明することを目的とする。破骨細胞を認識するモノクローナル抗体の1つを用いた免疫アフィニティークロマトグラフィーにより抗原の精製を行いSDS電気泳動後、LC-MASSを用いた質量分析を行った。候補蛋白質の1つとしてガレクチン3が検出されたが、ガレクチン3が抗原そのものであるのか、あるいは抗原に会合した分子であるのか、に関しては特定できなかった。ところで、ガレクチン3はMac2抗原として破骨細胞が発現することが報告されているが、その機能は分かっていなかった。特異的siRNAによりガレクチン3の遺伝子発現阻止を行い分化における機能を解析したが、破骨細胞分化への関与は認められなかった。そこでリコンビナントガレクチン3をRANKLに依存した破骨細胞分化系に添加したところ、破骨細胞の形成が顕著に阻害された。アジュバント関節炎ラットの系でも関節腔にガレクチン3を投与すると破骨細胞の形成が抑制され骨吸収が軽減されることがわかった。本研究より、ガレクチン3が破骨細胞の活性化を制御する重要な制御因子であることが強く示唆された。
