Research Abstract |
本研究では、当初、仙台ウイルス(HVJ)・エンベロープ・ベクターを用い、分泌マーカーcDNAと分泌関連タンパク質のsiRNAをラット顎下腺に導入し、唾液分泌機構のin vivo解析を計画した。しかし、予期に反し、このエンベロープ・ベクターには遺伝子導入活性がなかったため、計画変更を迫られた。そこで、培養細胞に、分泌マーカーとSNAREタンパク質を別の色の蛍光タンパク質として発現し、二波長全反射蛍光顕微鏡観察法により、構成的分泌を調節する分泌関連タンパク質の同定を試みており、現在、siRNAの効果から対象が徐々にしぼられてきている。
