| Research Abstract |
Loop-Mediated Isothermal Amplification法(以下、LAMP法とする)は、等温条件下で鎖置換反応を利用して目的の部位を増幅し、その際に生じる大量の副産物(ピロリン酸マグネシウム)が反応液を白く濁らせる状況を、目視あるいはリアルタイム濁度検出装置によって判定を可能とする検査法である。そこで、LAMP法によるMN式血液型判定について検討した。その結果、判定までの時間が約50分~55分で可能となり、また、Loopプライマーを添加すれば30分で判定可能とした。さらに、リアルタイムPCR法を用いてABO式およびMN式遺伝子型の同時検出を試み、約40分で同時検出を可能とした。
