Research Abstract

Ras-PI3K (phosphatidylinositol 3'-kinase)経路は、p110α(PIK3CA 遺伝子)、PTEN、K-Ras等の遺伝子発現異常により多くの癌で活性化され、細胞増殖に重要な役割を果たしている。我々は、PI3K のClass IA 触媒サブユニットp110α、β、δのどのアイソフォームが活性化されているのかを解析し、PIK3CA 変異株は、癌種によらず、常にp110αが優位に活性化していたこと、また、PTEN のみに変異を有する子宮体癌細胞株でもαが優位であるものがみられ、PI3K 経路の活性化におけるp110αの重要性を明らかにした。また、PIK3CA 変異が、他のRas-PI3K経路の遺伝子発現異常と浸潤癌特異的に共存している意義について解析し、PIK3CA変異はLate Event として、他の因子との共存によりRas-PI3K 経路をより強く活性化し、効率的に浸潤癌への悪性転化(Progression)を誘導していることを明らかとした。
