高解像度光学顕微鏡によるシナプス内部構造の検出

Research Abstract

中枢神経シナプスは神経細胞間の情報伝達の場であり、そこに局在する分子の分布を高い解像度で測定する事は、シナプスでの構造と機能の関連を理解する上できわめて重要である。中枢神経系の興奮性シナプスは、そのサイズが数ミクロンと小さいが、STORM顕微鏡を利用して高解像度の分子分布を知ることが出来れば、シナプス内部構造についての理解が格段に進展すると考えられる。この目的の為、前年度に引き続き以下の実験を行った。
1.STORM顕微鏡によるシナプス観察 対物レンズ型の全反射顕微鏡を使用して、一分子蛍光像を取得した。光スイッチとしてはPSD95-YFP,Homer-YFP,mGluR5-YFPを使用し、これらの蛋白質を海馬の分散培養神経細胞内にアデノウィルスを、用いて発現させた。水銀ランプの紫外光により光スイッチをオンにして、一分子に由来する蛍光像を取得し、その重心位置を推定した。全ての蛍光スポットが消退した後、再度紫外光による光スイッチの活性化を行い、このサイクルを数百回反復して一分子由来の蛍光像を積算した。シナプス後部に対応する構造をSTORM法により検出することが可能となった。
2.STORM法による分子位置推定法の評価 分子の位置を推定する方法として、三種類の計算方法を用いたプログラムを作成し、それぞれの方法における誤差を、シミュレーションにより解析した。ガウス分布を直接フィットすることにより中心位置を計算するプログラムを利用すると、最も精度の高い分子位置の推定が出来ることが示された。この計算方法を適用して、PSD95-YFPの分布をシナプス後部上で推定し、通常の蛍光顕微鏡法による形態との比較を行った。光学的なボケを除去した画像が得られ、通常の蛍光顕微鏡画像よりも限局した範囲にPSDの足場蛋白質が局在している可能性が示唆された。
