Research Abstract

当該研究の「研究の目的」及び「研究実施計画書」に沿って行った平成19～20年度の研究実績の概要を以下に記す。
糖を外殻に配置した糖デンドリマー型配位子とそのGd錯体の調製方法として次の5つのカテゴリーを組み合わせて、9パターン数十種類の新規なMRI造影剤を調製した。
カテゴリー:(a)鎖式/環式、(b)対称型/非対称型、(c)糖誘導体/非糖誘導体、(d)加水分解経路/非加水分解経路、(e)主鎖置換基/末端置換基
9パターン:(1-1)鎖式/対称/糖/非加水分解/末端、(1-2)鎖式/対称/非糖/非加水分解/末端、(1-3)鎖式/非対称/糖/非加水分解/末端、(1-4)鎖式/非対称/非糖/非加水分解/末端、(2)鎖式/対称/糖/加水分解/末端、(3)環式、(4)その他(主鎖置換、複合機能化、等)
In vitro評価:パターン(2)において縦緩和率(r1)30[s^<-1>・mM^<-1>]、パターン(1-3)においてr1=24[s^<-1>・mM^<-1>]を得た(目標値r1≒35[s^<-1>・mM^<-1>]の達成率70～85%)。また、パターン(1-2)において糖以外の基によるr1増強効果(例えば、フェニル基ではr1=11[s^<-1>・mM^<-1>])を確認した。
In vivo評価:従来品Gd-DTPAと比較して顕著な優位性は次の点であった。(1)血管造影について:パターン(2)では24時間、パターン(1-1)では2時間の十分な造影効果が見られた。(2)がん造影について:パターン(2)では極めて鮮明ながんの造影(濃度1/2)ができた。また、パターン(1-3)では初期がん(1.2mm程度のがん)造影(濃度1/8)に成功した(達成率80%)。
血管貯留性・生体内分子認識:パターン(2)ではin vivo評価により強い生体内分子認識を確認した。パターン(1-1)ではin vitro評価(二次元電気泳動、放射性^<153>Gd錯体)により弱いアルブミン認識を示した。分子設計ソフトもアルブミン-造影剤相互作用を支持した。
安全性試験:3種類のGd錯体に対して毒性の問題はなかった。
当該の基礎研究の達成度は80%程度であった。パターン(1)に対しては糖及びその他の機能性残基との複合化、パターン(2)に対しては造影剤の再現性、等の今後の課題が明らかになった。
