Research Abstract

核膜を標識するためにラミンBを、中心体の標識にセントリン2を、細胞膜の標識にβ-カテニン、E-カドヘリンを用いた積まず、これらのタンパク質のcDNAをRT-PCRにて取得し、そのcDNAの5'側または3'側に蛍光色素をコードするcDNAをin frameで連結し、標識タンパク質と蛍光色素の融合タンパク質をコードするcDNAを作製した。融合タンパク質のcDNAを発現ベクターにクローニングし、腫瘍細胞へ発現し、蛍光顕微鏡にて観察し、融合タンパク質が、予想された目的とする細胞内局在をとることを確認した。
このベクターを鋳型にRNA polymeraseを用いて転写反応を行い、RNAを作製した。さらにpoly Aを付加した後、精製し、注入用RNAとした。このRNAをマウス一細胞期胚に注入した。RNA注入後、48時間後に蛍光顕微鏡で観察し、期待される細胞内局在をとっているか、胚の発生は正常に進行しているかを判定した。さらにこの胚を固定し、各小器官に特有のタンパク質に対する抗体で、免疫染色を行い、発現させた融合タンパク質と共局在することを確認した。その結果、ラミンBおよびE-カドヘリンは、有用なツールであることが判明したが、中心体を標識するセントリン2は、発現が弱く明瞭な染色が得られなかった。
ラミンBおよびE-カドヘリンは、さらに継時的に観察をタイムラプス蛍光顕微鏡で行った。RNA注入12時間後より、30分ごとに96時間までの観察を行い。この間の蛍光強度の変化を調べたところ、いずれも96時間の間に蛍光強度は減衰するものの、観察は可能であることが判明した。
