Research Abstract

近位尿細管特異的に発現しているグルコーストランスポーターsglt2のプロモーター遺伝子領域と近位尿細管特異的にCre蛋白を発現するSglt2-Creマウスをフランスのグループより入手した。予備実験としてGFP蛋白を発現する事が可能なROSA26-tTA-GFP-loxPマウスと近位尿細管特異的プロモーターであるSglt2-Creマウスを交配し、近位尿細管特異的にGFP、tTAを発現するマウスを作製し、実際このプロモーターが目的蛋白質(予備実験ではGFP)を近位尿細管に発現させることが出来ることが確認されたので、このプロモーターを用いて腎臓特異的にヒトOATP-Rを過剰発現させたトランスジェニックラットを作製した。
hOATP-Rは、腎臓からのジゴキシン排泄に関与する事が知られている(Mikkaichi T et al. Proc Natl Acad Sci USA 101 : 3569-3574, 2004)。作製されたsglt2-hOATP-Rトランスジェニックラットにて、尿中ジゴキシン排泄について検討した。その結果、ヒトOATP-Rの腎特異的発現ラットであるTG(+)ラット(N=6)では、陰性対象であるTG(-)ラット(N=5)に比べて、優意に投与ジゴキシン量に対する尿中ジゴキシン排泄量の比の増加が確認でき、近位尿細管特異的に遺伝子を発現させる方法が樹立された。
