Research Abstract

家族性ALSの原因遺伝子ALS8 (VAPB)に着目してその機能解析と遺伝子変異による運動神経細胞死の機序を解明する研究を継続した。
目標1生理的UPRにおけるVAPBの果たす役割解明.
VAPBと結合し、機能的相互作用するFFAT motif蛋白質を同定する作業をFinlandのDr Olkkonenらとの共同研究で行ったが、同定にはいたらなかった。一方、VAPBがPERK経路に抑制的に係っており、P56S VAPBはこの機能が失われることを発見した。このことから、VAPBは少なくともUPRの2経路で生理的な役割を担っていることが明らかとなった。また、P56SVAPBにより、野生型VAPBの機能が抑制されるメカニズムを明らかにした。従って、自身は機能的にnullであるP56S-VAPBは野生型VAPBの機能を抑制することにより、VAPBの機能をおおむね抑制すると考えられた。
目標2P56S-VAPB誘導性運動神経細胞死機序の解明.
P56S-VAPBによるCapase-3の活性化を明瞭に示すことは成功しなかったが、P56SVAPBを発現させるとNSC34運動神経細胞がThapsigardin等の科学物質で誘導するUPRに対して脆弱となり、より強く細胞死が誘導されることを報告した(Suzuki et al.,2009)。P56SVAPBによるUPRの機能不全が最終的に運動神経細胞死に脆弱にさせると考えられる。
目標3:マウスを用いたALS8遺伝子異常によるALSモデル作成
a)P56S-VAPBトランスジェーニックマウスモデルの作成
P56S-VAPBトランスジェーニックマウスとVAPBに対するsiRNAを高発現させるトランスジェーニックマウスの作成を試みたが成功しなかった。
