Research Abstract

酸性糖であるシアル酸は、腫瘍マーカーであるシアリルTnやシアリルLe^x、シアリルLe^aなど、癌関連抗原に多く見いだされ、癌との深い関連性が指摘されてきた。われわれはこれまで、Neu4シアリダーゼがムチン糖鎖を水解するユニークな基質特異性を持つこと、長さの異なるisoformを有し、long formの細胞内主局在はミトコンドリアであり、他は細胞内膜に局在すること,大腸には、Neu4はほとんどshort formのみが存在するが、大腸癌ではNeu4発現が有意に低下することをみいだした。さらに、癌細胞にこのシアリダーゼを過剰発現すると、細胞の運動能や浸潤能を低下させるなど、悪性形質が低下する傾向を示すが、遺伝子のノックダウンを行うと、悪性形質が増強することがわかった。
20年度は、NEU4によるこれらの現象の分子機構の一端を明らかにした。シアリルLe^xやシアリルLe^aを高発現している大腸癌細胞においてNEU4の発現が亢進すると、E-セレクチンを介するシグナリングが阻害を受けた。その下流にあるERKやp38,Hsp27が不活化を受け、この結果、細胞運動能や増殖能が低下することがわかった。癌細胞ではNEU4発現が低いために、シアリルLe^xやシアリルLe^aの高発現が起こり、その結果、浸潤や運動能が亢進していることが強く示唆された。今後はNEU4のノックダウン等によって、この現象の確認実験を行なう。
